Q5 High-Fidelity DNA Polymerase sets a new standard for both fidelity and robust performance. With the highest fidelity amplification available (~280 times higher than Taq), Q5 DNA Polymerase results in ultra-low error rates. Q5 DNA Polymerase is composed of a novel polymerase that is fused to the processivity-enhancing Sso7d DNA binding domain, improving speed, fidelity and reliability of performance. This kit contains a sufficient supply of master mix and nuclease-free water to perform 50 polymerase chain reactions or 200 reactions. For added convenience, gel loading dye, and a Quick-Load DNA LAdder are also included in this kit.
New England Biolabs, Inc. LongAmp Taq PCR Kit – 100 reactions (50 µl vol)
The LongAmp Taq PCR Kit features LongAmp Taq DNA Polymerase, which is a unique blend of Taq and Deep Vent DNA Polymerases. The 35 exonuclease activity of Deep Vent Polymerase increases the fidelity and robust amplification of Taq DNA Polymerase. LongAmp Taq DNA Polymerase offers two fold higher fidelity than Taq Polymerase alone. The LongAmp Taq PCR Kit is supplied with LongAmp Taq Reaction Buffer, which is optimized to give highest PCR yields up to 30 kb. The kit can generate a wide range of PCR products, up to 30 kb from plasmid or from human genomic DNA.
New England Biolabs, Inc. PreCR Repair Mix – 150 reactions
The PreCR Repair Mix is an enzyme cocktail formulated to repair damaged template DNA prior to its use in the polymerase chain reaction (PCR), microarrays or other DNA technologies. PreCR is active on a broad range of DNA damages, including those that block PCR (e.g. apurinic/apyrimidinic sites, thymine dimers, nicks and gaps) and those that are mutagenic (e.g. deaminated cytosine and 8-oxo-guanine). In addition, it will remove a variety of moieties from the 3end of DNA leaving a hydroxyl group. The PreCR Repair Mix will not repair all damages that inhibit/interfere with PCR. For example, it will not repair 8-oxo-7,8-dihydro-2'deoxyadenosines or fragmented DNA. In fact, the ligase present in the mix is very active at sealing nicks in DNA but does not ligate blunt ends or nicks near a mismatch effectively. This limits the possibility of chimeric gene formation as a product of ligation. The PreCR Repair Mix can be used in conjunction with any thermophilic polymerase.

Phusion High-Fidelity DNA Polymerase offers both high-fidelity and robust performance, and thus can be used for all PCR applifications. Its unique structure, a novel Pyrococcus-like enzyme fused with a processivity-enhancing domain, increases fidelity and speed. Phusion DNA Polymerase is an ideal choice for cloning and can be used for long or difficult amplicons. With an error rate >50-fold lower than of Taq DNA Polymerase and 6-fold lower than that of Pyrococcus furiosus DNA Polymerase, Phusion is one of the most accurate thermostable polymerases available. Phusion DNA Polymerase possesses 5' to 3' polymerase activity, 3' to 5' exonuclease activity and will generate blunt-ended products. The Phusion High-Fidelity PCR Kit contains a sufficient supply of Phusion High-Fidelity DNA Polymerase, Phusion HF and GC Buffers, deoxynucleotides, magnesium chloride, DMSO, and DNA size standard to perform 50 reactions or 200 reactions.
New England Biolabs, Inc. OneTaq Hot Start DNA Polymerase – 200 units
OneTaq Hot Start DNA Polymerase is an optimized blend of Taq and Deep Vent DNA polymerases combined with an aptamer-based inhibitor. The 3' to 5' exonuclease activity of Deep Vent DNA Polymerase increases the fidelity and robust amplification of Taq DNA Polymerase and the hot start formulation combines convenience with decreased interference from primer dimers and secondary products. The OneTaq Reaction Buffers and High GC Enhancer have been formulated for robust yields, regardless of a template's GC content. The inhibitor binds reversibly, blocking polymerase activity at temperatures below 45C. The polymerase is activated during normal cycling conditions, allowing reactions to be set up at room temperature. OneTaq Hot Start DNA Polymerase does not require a separate high temperature incubation step to activate the enzyme and can be used in typical Taq-based cycling protocols. It is supplied with two 5X buffers: (Standard and GC), as well as a High GC Enhancer solution.

The Q5 High-Fidelity 2X Master Mix offers robust, high-fidelity performance in a convenient master mix format. The Q5 High- Fidelity 2X Master Mix features a high-fidelity, thermostable DNA polymerase with 3' to 5' exonuclease activity, fused to a processivity-enhancing Sso7d domain to support robust DNA amplification. With an error rate ~280-fold lower than that of Taq DNA Polymerase, Q5 High-Fidelity DNA Polymerase is ideal for cloning and can be used for long or difficult amplicons. The convenient master mix formulation is supplied at a 2X concentration. The mix contains dNTPs, Mg++ and a proprietary broad-use buffer requiring only the addition of primers and DNA template for robust amplification regardless of GC content. When used at the recommended 1X final concentration, the Q5 High-Fidelity Master Mix contains 2 mM Mg++. Q5 High-Fidelity DNA Polymerase is unlike typical, lower fidelity PCR enzymes.

Deep Vent DNA Polymerase was the second high-fidelity thermophilic DNA polymerase available from New England Biolabs. The fidelity of Deep Vent DNA Polymerase is derived in part from an integral 3 5 proofreading exonuclease activity. Deep Vent is even more stable than Vent at temperatures of 95 to 100C.

OneTaq Hot Start Quick-Load 2X Master Mix with Standard Buffer is an optimized, ready-to-use blend of Taq and Deep Vent DNA Polymerases combined with an aptamer-based inhibitor. This enzyme blend is ideally suited to routine PCR applications from a variety of templates, including pure DNA solutions, bacterial colonies and cDNA products. The 3' to 5' exonuclease activity of Deep Vent DNA Polymerase increases the fidelity and robust amplification of Taq DNA Polymerase. The hot start nature of the enzyme offers convenience with decreased interference from primer dimers and secondary products. The convenient Quick-Load master mix formulation contains dNTPs, MgCl2, buffer components and stabilizers as well as two commonly used tracking dyes for DNA gels. On a 1% agarose gel in 1X TBE, Xylene Cyanol FF migrates at ~4 kb and Tartrazine migrates at ~10 bp. Both dyes are present in concentrations that do not mask co-migrating DNA bands
