Deep Vent DNA Polymerase was the second high-fidelity thermophilic DNA polymerase available from New England Biolabs. The fidelity of Deep Vent DNA Polymerase is derived in part from an integral 3 5 proofreading exonuclease activity. Deep Vent is even more stable than Vent at temperatures of 95 to 100C.

NEBuilder HiFi DNA Assembly Master Mix was developed to improve the efficiency and accuracy of DNA assembly. It allows for seamless assembly of multiple DNA fragments, regardless of fragment length or end compatibility. Has utility in synbio community, as well as those interested in one-step cloning of multiple fragments due to its ease of use, flexibility and simple MM format. The rxn includes different enzymes that work together in the same buffer: The exonuclease creates single-stranded 3' overhangs that facilitate the annealing of fragments that share complementarity at one end (the overlap region): a polymerase fills in gaps within each annealed fragment; DNA ligase seals nicks in the assembled DNA. New England Biolabs, Inc. Faustovirus Capping Enzyme - 500u
Faustovirus Capping Enzyme (FCE) catalyzes the addition of N7-methylguanosine cap (m7G) to the 5 prime end of triphosphorylated and diphosphorylated transcripts producing Cap-0 RNA. FCE is a single-subunit enzyme that combines the three activities necessary to produce the Cap-0 structure- triphosphatase guanylyltransferase and (guanine-N7)-methyltransferase. Installation of Cap-0 is a key step in eukaryotic mRNA maturation along with 2-O-methylation at position 1 (Cap-1) and polyadenosine (poly(A)) tailing. The Cap-0 structure also promotes RNA stability and prevents inadvertent activation of innate immune responses triggered by triphosphorylated RNA. FCE retains significant capping activity at low temperatures and tolerates reaction temperatures up to 55C. In many cases 1 uL of FCE (25 units) can cap over 100 ug of RNA in 1 hour at 37C. GTP and S-adenosylmethionine (SAM) are required for capping activity and are included with the enzyme.

DNA Polymerase I, Large (Klenow) Fragment is a proteolytic product of E. coli DNA Polymerase I which retains polymerization and 3' to 5' exonuclease activity, but has lost 5' to 3' exonuclease activity. Klenow retains the polymerization fidelity of the holoenzyme without degrading 5' termini.

RecJf is a single-stranded DNA specific exonuclease that catalyzes the removal of deoxy-nucleotide monophosphates from DNA in the 5' to 3' direction. RecJf is a recombinant fusion protein of RecJ and maltose binding protein (MBP). It has the same enzymatic properties as wild-type RecJ. Fusion to MBP enhances RecJf solubility. When DNA containing a 22 base 5' extension is used as a substrate for RecJf, the resulting products are a mixture of DNA fragments that have blunt-ends, 5' extensions (1-5 nucleotides) and recessed 5' ends (1-8 nucleotides). RecJf does not require a 5' phosphate.
