Description
The Human Vascular Cell Adhesion Molecula-1 (Hu VCAM-1) ELISA quantitates Hu VCAM-1 in human serum, plasma, amniotic fluid, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu VcAM-1. Principle of the method The Human VCAM-1 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. VCAM-1 (vascular cell adhesion protein 1, CD106, INCAM-110) is a cell surface sialoglycoprotein expressed by cytokine activated endothelium. VCAM-1 has a number of functions including the regulation of leukocyte migration, leukocyte-endothelial cell adhesion and signal transduction and may play a role in a number of inflammatory diseases. VCAM-1 is distributed across non-leukocyte and leukocyte cells. VCAM-1 expression is reportedly correlated with adhesion of melanoma, Burkitt lymphoma, osteosarcoma, and kidney carcinoma but not colon carcinoma cells to endothelial cells which may determine the site of tumor metastasis. VCAM-1 is a member of the Ig superfamily of adhesion molecules, is expressed at high levels on cytokine stimulated vascular endothelial cells, and at minimal levels on unstimulated endothelial cells. VCAM-1 is also present on follicular and inter-follicular dendritic cells of lymph nodes, myoblasts, and some macrophages. VCAM-1 serves as a ligand for leukocyte integrin alpha-beta (VLA-4 or CD49d/CD29) and mediates cell adhesion of leukocytes to activated endothelium. VCAM-1 plays a role in tumor metastasis, and may also be involved in the development of atherosclerosis and rheumatoid arthritis. Two alternatively spliced transcripts encoding different isoforms of VCAM-1 have been described.Specifications
