Accelerated screening for protein stability as a function of pH, ionic strength, and concentration. The PROTEOSTAT Thermal shift stability assay kit includes a fluorescent dye which detects protein aggregation, so it can be used to monitor protein stability under systematic thermal stress conditions. From the thermal shift assay, a temperature at which the bulk of the protein becomes aggregated can readily be identified. The aggregation temperature is an indicator of protein stability and can be used to optimize conditions that minimize protein aggregation as well as to identify ligands that bind and confer structural stability to a protein of interest. The EFLUXX-ID Multidrug Resistance Assay Kit allows for functional detection of all three clinically relevant ABC transporter proteins: MDR1 (p-glycoprotein), MRP1/2, and BCRP. The assay uses a hydrophobic, non-fluorescent compound that readily penetrates the cell membrane, where it is hydrolyzed to a hydrophilic fluorescent dye by intracellular esterases. Unless the EFLUXX-ID dye is pumped out of the cell, the esterase cleaved dye is trapped inside the cell. Thus, cells exhibiting drug resistance will have diminished fluorescence. EFLUXX-ID assay is the only available kit for the simultaneous monitoring of all three major ABC reporter proteins with the ability to profile individual pump activity. The kit offers a set of six different MAbs specific for serine phosphorylation sites. Phosphorylation patterns in a given cell extract may differ when probed with different antibodies due to sequence specificity. Recognition therefore depends on two criteria: phosphorylation and surrounding aa motif. If one of the two criteria is not fulfilled, the antibody will not detect the phosphorylation site. Specificity has been determined by epitope mapping using degenerated phosphopeptide libraries. Clone 16B4 strongly interacts with pSK and pSP motifs, thus specifically recognizes substrates of MAP/SAP Kinases and CDC kinases.Clones 1C8, 4A3, 4A9, 4H4 and 7F12 interact with epitopes containing phosphoserine in the context of positive or neutral aa (e.g. substrates of PKA, PKB, PKC, PKG etc.) Though revealing a similar pattern in epitope mapping using phosphopeptide libraries, these antibodies interact with different sets of phosphoproteins in whole cell lysates. The assay procedure has two steps. First, the COLOR DE LYS substrate which comprises an acetylated lysine side chain, is incubated with a sample containing HDAC activity . Deacetylation of the substrate sensitizes the substrate so that, in the second step, mixing with the COLOR DE LYS developer causes an increase in yellow color intensity, and absorption at 405 nm.
