Tumor necrosis factor-α (TNF-α) is a pro-inflammatory cytokine that activates NF-κB, MAPK, and PI3K/AKT pathways. Activated T cells and macrophages are the primary producers of TNF-α in response to inflammation and infectious conditions. Many other cell types have been shown to produce TNF-α, among them B cells, NK cells, mast cells, neutrophils, dendritic cells, microglia, endothelial cells, smooth muscle cells, cardiomyocytes, and fibroblasts. TNF-α has cytotoxic effects on cancer cells in vitro by stimulating anti-tumor immunosuppressive responses. TNF-α stimulates expression of E- and P-selectins, thus facilitating adhesion of neutrophils, monocytes, and memory T cells to activated platelets and endothelial cells (Zelová & Hosek). Other effects of TNF-α include vasodilatation and edema formation.

The SARS-CoV-2 (Spike Protein) Peptide Pool is provided as two lyophilized mixtures (subpools) from the SARS-CoV-2 spike protein. Each subpool contains 158 peptides, for a total of 316 peptides. The virus attaches to the cell membrane of the host through the interaction between spike protein and angiotensin-converting enzyme 2 (ACE2) receptor, and the spike protein plays a critical role in viral entry. The subpools consist of 15-mer peptides with 11-amino-acid overlaps that cover amino acids 1 - 1273 on the spike protein.

LysoBrite™ dyes are fluorescent dyes that selectively stain lysosomes and other acidic organelles. Due to their hydrophobic nature, LysoBrite™ dyes permeate live cells and selectively accumulate in the lysosomes via the lysosome’s pH gradient. The fluorescence property of these dyes significantly increases within the lysosome’s acidic environment; this fluorescence can be measured using flow cytometry and fluorescence imaging. Due to their selective fluorescence inside lysosomes, LysoBrite™ dyes provide low background and high contrast during imaging. They can be used with proliferating and non-proliferating cells, and are suitable for suspension and adherent cells.

Propidium Iodide (PI) is a red-fluorescent cell viability dye which is excluded from live cells with intact membranes, but penetrates dead or damaged cells and binds to DNA and RNA by intercalating between the bases. It is widely used as a counterstain to differentiate and exclude non-viable cells in flow cytometric analyses, and can be excited using blue (488 nm), green (532 nm), or yellow-green (561 nm) laser lines, with detection in the FL2 or FL3 channels. PI is used in DNA fluorescence imaging applications to discriminate early and late stages of apoptosis, to study cell‐mediated cytotoxicity, and for chromosome analysis. It is also commonly used in quantitative DNA assays.
STEMCELL Technologies ReproRNA™-OKSGM, Size: 12 µg
ReproRNA™-OKSGM is a single-stranded RNA replicon vector that contains five reprogramming factors: OCT4, KLF-4, SOX2, GLIS1, and c-MYC, as well as a puromycin-resistance gene (Figure 1). This RNA vector reprograms somatic cells, such as fibroblasts, into induced pluripotent stem (iPS) cells with high efficiency and only requires a single transfection step (Figure 2). When used together with ReproTeSR™ reprogramming medium, the generation of iPS cell colonies can be achieved under feeder-free conditions with superior colony morphology and similar reprogramming efficiency to feeder-based systems (Figures 3 - 4). ReproRNA™-derived iPS cell colonies also express markers of undifferentiated cells and retain a normal karyotype (Figures 5-6). Subsequently, iPS cells generated with ReproRNA™-OKSGM can be maintained in TeSR™ maintenance media (mTeSR™1, TeSR™2, or TeSR™-E8™)
