Description
Streamline therapeutic antibody discovery campaigns with Zenon Alexa Fluor Plus Labeling Reagents to screen for antibody binding. These reagents provide rapid, scalable antibody labeling and give a bright fluorescence-based indication of antibody binding when analyzed by flow cytometry, fluorescence microscopy, or high-content screening.
An effective cell-based screening model is critical for the success of therapeutic antibody discovery campaigns. This serves as an important filter in the early stages of drug development. Identifying antibodies with optimal binding profiles accelerates candidate selection and helps minimize false positives. The new Zenon Alexa Fluor Plus Labeling Reagents are prelabeled Fab fragments directed against the Fc portion of IgG antibodies. They are improved in three ways: superior pan-reactive Fab fragments, optimized bioconjugation, and next-generation fluorophores. These combine to provide superior signal to background, providing more confidence in screening results.
The Zenon Alexa Fluor Plus IgG Labeling Reagents contain sufficient material to label one to four 96-well plates, depending on the Zenon labeling reagent concentration used. Formation of the Fab–antibody complex occurs in less than 5 minutes. Only a small amount of starting material is needed, and the method is optimized for efficient labeling of antibodies in serum, ascites fluid, or hybridoma suspensions. The insights provided by these approaches have the potential to accelerate biotherapeutic lead generation and development.
Advantages of Zenon Alexa Fluor Plus IgG Labeling Reagents include:
• Simplicity—No reagent purification is required
• Accuracy—No wash steps or quenchers are needed, maximizing throughput
• Speed—Labeled antibodies are typically ready to use in 5 minutes
• Specificity—Dyes are designed for the specific detection of antibody binding
Preserve primary antibody function and affinity
Reactive dye labeling of primary antibodies can have unpredictable and undesirable outcomes. One of these is reduced binding affinity due to label addition in the binding pocket. The Zenon antibody labeling approach, which is targeted to the Fc tail, resolves this potential issue.
