Preparation: Produced from sera of goats immunized with highly pure Recombinant Human TPO. Anti-Human TPO-specific antibody was purified by affinity chromatography employing an immobilized Human TPO matrix. Sandwich ELISA: To detect Human TPO by sandwich ELISA (using 100 μL/well antibody solution) a concentration of 0.5-2.0 μg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human TPO (500-P49GBt) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of Recombinant Human TPO. Western Blot: To detect Human TPO by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagents the detection limit for Recombinant Human TPO is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.
Thrombopoietin (TPO, THPO, MPL ligand, megakaryocyte colony-stimulating factor) is responsible for the generation of platelets through its action on hematopoietic cells committed to the megakaryocyte lineage. Binding of thrombopoietin to its receptor, MPL (CD110), initiates a signaling cascade via JAK/STAT, MAPK, and PI3K/AKT signaling pathways that leads to the proliferation and differentiation of megakaryocytes and, μgtimately, to the production of platelets (thrombopoiesis). Thrombopoietin is constitutively produced by the liver and has a predicted molecular weight of 38 kDa.Specifications
