Identify yeast and anaerobic microorganisms, primarily Candida albicans and Clostridium difficile, using Thermo Scientific™ Remel™ PRO Kit. PRO Kit is recommended for use in qualitative procedures for detection of the enzyme L-Proline aminopeptidase in yeast and bacterial cells. The PRO Kit may be used as a single colony screening test to aid in the identification of Candida albicans and Clostridium difficile. The PRO disk is 99% positive when testing C. difficile from anaerobic cultures. It is useful in differentiating other Gram-positive and Gram-negative bacilli.

Use PRO Kit for identification of Candida albicans and Clostridium difficile. C. albicans is the yeast pathogen most frequently isolated from clinical specimens1. In 1990, Perry et al. evaluated a rapid test to aid in the identification of C. albicans based on colorimetric detection of the enzyme, L-proline aminopeptidase2. C. difficile is recognized as the major cause of antibiotic-associated pseudomembranous colitis and a frequent cause of hospital-acquired diarrhoea. Fedorko and Williams reported detection of L-Proline aminopeptidase is useful for identification of C. difficile because it is one of the few biochemical tests for which this organism is positive3.

The PRO disk contains the substrate L-proline-β-naphthylamide. This substrate is hydrolyzed by an aminopeptidase releasing β-naphthylamine. After addition of Cinnamaldehyde Reagent, β-naphthylamine reacts with ρ- dimethyl-aminocinnamaldehyde to form a red-colored complex.
