Description
MabCaptureC™ RoboColumn™ are small chromatography columns used for fully automated and parallel chromatographic separations on robotic liquid handling workstations.
These pre-packed columns are ready to connect and have the following features:
- Packed with MabCaptureC™ Affinity Matrix, offering high dynamic binding capacity and high resolution over a wide range of flow rates
- Direct connection to standard chromatography systems
The 1 mL columns can be used for quick screenings of application feasibility and lab scale purification on a convenient and easy-to-use pre-packed column. The 10-cm bed height of the 5 mL column allows initial process development at a bench scale.
The pre-packed MabCaptureC™ Affinity Matrix contains engineered rProtein A that binds at the CH2-CH3 interface of the Fc-region of IgGs and shows cross binding to the VH3 domain of IgGs.
Features of MabCaptureC™ Affinity Matrix include:
- High dynamic mAb binding capacity of > 50 g/L at 4.8 min residence time
- Purification of mAbs and a subpopulation of scFv's and Fab fragments containing VH3 binding domains
- Clean and sanitize with standard sodium hydroxide agents for chromatography products
- Excellent scalability
- Non-animal derived
Main resin characteristics:
- Matrix: agarose-based, epoxide activated
- Average particle size: 75 ± 10 μm
- Ligand: recombinant C domain of Protein A
- Ligand coupling method: epoxide coupling
- Binding capacity: >55 g IgG/L resin at 5 min residence time and 10% breakthrough; >75 g IgG/L resin at 10 min. residence time and 10% breakthrough
- Elution conditions: 0.1 M glycine pH 3.0, or 20–100 mM sodium citrate or acetate, pH 3.0
- Cleaning conditions: 0.2 M sodium hydroxide for 15 minutes every cycle
- Column dimensions: 0.8 x 2 cm (1 mL), 0.8 x 10 cm (5 mL)
- Operating pressure: ≤ 3 bar
- Formulation buffer: 20% (v/v) ethanol
