Use dickkopf-related protein 1 (DKK-1) to drive cardiac and retinal differentiation from induced pluripotent stem (iPS) cells (Lian et al. Proc Natl Acad Sci USA 2012). DKK-1 is a secreted protein that inhibits the canonical WNT pathway by competitive binding to low-density lipoprotein receptors (LRP)-5 and 6 (Niehrs. Oncogene 2006). This protein regulates embryonic development (Lieven O et al. Dev Biol 2010) and has been shown to support the generation of myeloid-derived suppressor cells (MDSCs) (D’Amico et al. J Exp Med 2016). DKK-1 from thrombocytes also induces Th2 cell polarization and potentiates Th2 cell cytokine expression (Chae et al. Immunity 2016). Human recombinant DKK-1 from STEMCELL contains a His-residue tag at the carboxyl end of the polypeptide chain and comes lyophilized with ≥ 95% purity EC50 4 μg/mL endotoxin levels verified 0.2 EU/μg protein.
