Description

Alexa Fluor antibody labeling kits provide a convenient means to label antibodies or proteins (>40 kDa) with as many as nine different colored dyes. Conjugated antibodies produce lower background fluorescence with less nonspecific binding and are ideal for multiplexing in a variety of applications. Each kit contains sufficient dye to conjugate 100 µg per reaction and enough reagent for five reactions. In as little as 90 minutes, your samples can be conjugated and ready to use.

Using the fluorescent dyes provided in our Alexa Fluor antibody labeling kits allows multiple primary antibodies of the same isotype, or antibodies derived from the same species, to be used in the same experiment by directly labeling them with compatible fluorophores. Conjugates are ideal for multiple applications including flow cytometry, fluorescent microscopy techniques, immunocytochemistry, immunohistochemistry, primary detection, ELISA, indirect FISH, and more. Each kit contains all required reagents and fluorophores to perform five separate labeling reactions as well as to purify the resulting conjugates. These kits are compatible with each other, easy to use, and make multiplexing a reality in various applications requiring imaging.

• Label up to 100 µg of protein per reaction (five reactions per kit)
• Label proteins greater than 40 kDa
• Includes convenient spin filters
• Stabilizing proteins such as BSA should be removed from the sample before labeling
• Includes detailed instructions for determining degree of labeling (DOL)

Yellow-fluorescent Alexa Fluor 532 —excitation and emission maxima of 531/554 nm; excited using a 532-nm Nd:YAG laser line and detected under standard rhodamine 6G filters
Orange-fluorescent Alexa Fluor 546—excitation and emission maxima of 558/573 nm; excited using a 543-nm He-Ne laser line and detected under standard TRITC/Cy3 filters
Orange-fluorescent Alexa Fluor 555—excitation and emission maxima of 553/566 nm; excited using a 543-nm He-Ne laser line and detected under standard TRITC/Cy3 filters
Orange-red-fluorescent Alexa Fluor 568—excitation and emission maxima of 579/603 nm; excited using a 568-nm Kr laser line and detected under standard Rhodamine Red/Cy3.5 filters
Red-fluorescent Alexa Fluor 594—excitation and emission maxima of 590/616 nm, excited using a 594-nm Kr or He-Ne laser line and detected under standard Texas Red filters
Far-red-fluorescent Alexa Fluor 647—excitation and emission maxima of 650/668 nm, excited using a 633- or 635-nm Kr or He-Ne laser line and detected under standard APC/Cy5 filters
Near-IR-fluorescent Alexa Fluor 680—excitation and emission maxima of 680/700 nm
Near-IR-fluorescent Alexa Fluor 790—excitation and emission maxima of 784/814 nm

Compared to traditional dyes, Alexa Fluor dyes are brighter, more photostable, and more pH resistant between pH 4 and 10.
